Yes, it definitely can. Being cynical can become very negative.

Of course they can be a turnoff, yet it depends on who they are expressing their cynicism with. Reason: misery does love company. So, if the cynic is expressing there's no need in applying for that cool job at that cool company as they only hire Ivy League grads to someone who is depressed and just lost their job and has been applying everywhere for 6 months, this depressed person will not be turned off by the cynic and may enjoy venting about how the system is rigged to keep you from moving up a class. However, if someone who enjoys life, recently lost their job due to cutbacks and is trying like heck to get another job and feels confident they will find a job soon because they have a great skill set, has a cynic tell them they shouldnt apply for that cool job at that cool company as they only hire Ivy League grads, this optimistic person will feel uncomfortable with the cynics comments as they will see them as a mismatch to their hopeful outlook. If the cynic keeps on with this, the happy person is likely to lose interest and stop talking to them period.
