Go to sheldonbrown.com and look at his wheel building instructions. Give it a try. You won't get a good wheel on the first try, but it's a fun thing to learn how to do.

Ever heard about the expensive French restaurant? As the old saying goes, "If you have to ask the price, you can't afford to eat here." SAME THING! If you have to ask here (on Yahoo of all places) it's beyond your skill level.
Lacing a wheel takes YEARS of practice + all the special tools. Unless you plan on making this your career, pay the bike shop to do it. So you'll be out $80 bucks or so. You'll spend that much or more for aspirin curing headaches trying to do it yourself.

Let's start this question off simple, do you have a truing stand and maybe even a dishing attachment? There's lots of YouTube videos that teach you how to lace wheels.
