What is a stroke?

She was in the light of the fading moon and the graceful stroke of clouds passing over slowly lightening the horizon.

    It means they had an impact on the scenary that she was observing changing completely the atmosphere.

    a brief passing by, as in stroke of someone's hair or stroking the water to propel a canoe.  Or the passing of a brush while painting, which is somewhat the metaphorical idea intended by the wording.  it is a picturesque choice of words that is not exactly literal.  the sky is like someone had painted it with wisps of passing clouds.

    The "stroke" in this example indicates that the way the clouds were passing over were seen in a quick or flirting manner. It would be similar to say a dollop when cooking. You would add a dollop of cream to a sauce. It is not an exact measure, but means "just a little bit".

    Clouds don't lighten anything directly, they reflect or scatter light from another source. The stroke is referring to a paint brush stroke in a line.
