The best way is to get plenty of rest before a long drive - and if you find yourself drowsing off, pull over and get some rest.
Sometimes you just need to pull over, get out and stretch - maybe do some light exercise to get your blood pumping.
Caffeine and sugar are good for a short-term pick-me-up but you can end up more exhausted when you come down.
Cranking up some music and singing along sometimes helps too.

Not possible. You might as well ask how to prevent alcohol or heroin from affecting your driving, because the answer is the same. You don't.
Fatigue is just like drugs or alcohol, only not nearly as fun. But just like drugs and alcohol (or both), it's really not good for driving. When you're suffering from fatigue, your reaction time is slower, your senses are dulled and your mind can't focus on the task at hand.
The only way to prevent fatigue from affecting your driving is to not drive when fatigued. There is no other way.

Best thing is to take a break every two to four hours. Like walk around for a half hour in the rest area, breath some fresh air, stretch, drink something, water or caffeinated beverage.
That's about it. You just need to allow some extra time for rest breaks and or delays like car accidents that shut down the roads.
