Of course they can. Comedians can normally walk on a stage with a prepared set no matter what is going on internally and deliver it with real professionalism. Maria Bamford talks about just this in many of her shows. How she deals with mental illness and being a comedian. Patton Oswalt. Many perform despite being a mess off-stage. They perform to deal with it, a lot of the time.

Humor is a defense mechanism that comes from tragedy. There's no getting around it.

Its a tough part of their job.  In fact, Lou Costello went on stage and cracked jokes to an audience after his 3 year old son had died in a swimming pool.

Who could feel sad or happy all of the time? Of course they can feel sad while still performing. It doesn't have to mean that they have to be funny to feel sad vice versa, etc.

No. If a comedian ever feels the slightest bit of sadness, they lose their job.

Being a comedian is their job and for most of them the material is rehearsed and memorized much like an actor will memorize a script,, sad or not, when the curtain goes up and the lights go on they have a job to do,, so they put on the funny face and do their job.
