Could you carry Lake Ontario up a flight of stairs?

Like if you spent your whole life on it, breaking to eat and sleep obviously. Could you carry Lake Ontario, bucket by bucket, by hand, up an average flight of stairs (12 steps)?

With enough buckets and faith, anything is possible.

Sure, one bucket at a time, but first you would need to come up with a serum for immortality.

The volume is 1.6*10^12 cubic meters. A bucket contains about 5*10^-3 cubic meters. So that would be 3.3*10^14 buckets. Assuming you could carry 1000 buckets a day for 40 years, that would be 14 million buckets. You would need 23 million people to do the same to achieve your goal.
