If you are adding a ' then the word is possessive.

eg The 5 countries' ambassadors arrived at the meeting.

To be plural it would not have a '

eg The ambassadors of the 5 countries arrived at the meeting.

    It is both.

Countries is the plural of country.

The adding of an possessive apostrophe makes the word possessive.

These countries have vast oil reserves.

This country has a big GNP.

It is these countries' right to have free trade agreements with other countries.

    That country's GNP is large.

    "Countries" is plural of country.

"Countries'" is possessive of the plural. e.g. "The countries' border" could mean the border between two countries.
