What's the best way to get from Frankfurt to Berlin?

I am going on holiday to Frankfurt next month and want to go to berlin for a day. Is it cheap to get a train there and stay at a hotel for a night?

I used to live in Germany and travelled a lot there. It's quite a distance (about 350 miles) from Frankfurt to Berlin but worth the travel if you are going to stay over night.
As an alternative Air Berlin www.airberlin.com fly between the two cities and I have to say offer a great service.
Both these sites are available in English.
You will find train travel at weekends cheaper especially if you go on a slower train but for what it's worth fly.

Highspeed trains (the "ICE") are quite expensive when you book to late and otherwise the travel would take serveral hours. You can visit www.bahn.de (language selection right on top) for fares and scedules.

Another possibility would be to fly, due to the fact that there are several low cost carriers in Germany. You should check the websites of "airberlin", "TUIfly" and "germanwings" and you could have a look at Lufthansa too, they have special conditions often.

It will take you approx. 1/2 day each way to and from FRA to Berlin, with both Train or Plane.

Trains travel from inner-city to inner-city and thus are quicker to access but take longer. If you consider the time it takes to travel to and from the airports and the time you have to be there early for check-in, the difference is not significantly.

If you have only one day to visit Berlin, consider taking an overnight (slow) train over the ICE (fast). The cabins are quiet comfortable with coffee delivered in the morning, you save the hotel costs and it's cheaper than the fast ICE (and at night the slow speed doesn't matter).

There is e.g. the City Night Line (CNL 1259) train for 91 Euros non-stop. If you book outside Germany, check for tourist discounts.

Some scenery will be missed along the route by night train but during my last trip I noticed a frequent site of wind turbines that (despite their great environmental purpose) put a damper onto my sight seeing experience.

If you meant cheapest way, you will have to plan well ahead. The train costs only €29 if booked well in advance and online. You will have to print the ticket yourself. Spontaneous travel by train will set you back over €100 (one way) which is in fact also the cost for a cheap hotel room in downtown Berlin, except hostels. Cheaper hotels are located at the outskirtsof town but hard to get to.
Flying can be cheaper if booked early.
The cheapest way to get to Berlin is perhaps sharing a car (www.mitfahrzentrale.de) either as driver (of a rental car) or as passenger.

Not sure why the very correct answers below have attracted so much negative feedback from a bunch of muppets, but I can assure you the guys-girls before me have told you all you needed to know about plane or train options and I just want to repeat that it is definitely a worthy trip to take if you stay overnight!
